When excitation occurs at 745 nm, the fluorescence from tissues is seen in the red, and when excited at 488 nm, the fluorescence from tissues is seen in the green. Because of the high absorbency of the 780 nm dye FITC at the 488 nm wavelength, the tissue fluorescence at this wavelength is suppressed.
In comparison to 745 nm excitation, 488 nm excitation gives a lower level of fluorescence from the normal and diseased tissues. At 488 nm, the ratio of the fluorescence from diseased tissues to the background is approximately 0.5 to 1:1. However, at 745 nm, the fluorescence from diseased tissue is reduced to about 3 to 5:1.
